Does direction of pipe inserts really matter for short run?

I am in one of the last stages of my basement remodel. I need to run about a 12 ft run of a return from the trunk to the bottom of floor. My newbie to hvac question is does it really matter if the inserts of the 6" pipe go into the opposite direction of air flow? It seems all the connections I got were made for a supply run. Hope this question is clear enough on what i mean.

It certainly doesn't make any difference on the supply runs.
All connections should be sealed with mastic or appropriate tape.
